蛋白激酶CK2β亚基在肝癌中上调表达及其对肝癌细胞增殖和迁移的影响 被引量:3

High Expression of β Subunit of Protein Kinase CK2 and the Effect to the Developing and Migration in HCC Cell Lines

摘要 蛋白激酶CK2,曾被称为酪蛋白激酶Ⅱ,是一种在真核细胞中普遍存在的信使非依赖性丝/苏氨酸蛋白激酶,高度保守且功能多样,在细胞的生长、增殖、凋亡以及生物节律的调控中均发挥重要的作用.本文主要研究CK2β亚基在肝癌中的表达变化及其与肝癌发生和迁移的关系.用Real-time PCR方法检测了该基因在91对肝癌组织样品中的表达情况,发现其表达在69例肝癌组织中相对其癌旁组织表达上调(占总样本数的75.8%).对75对肝癌与癌旁组织样品的免疫组化分析显示,CK2β亚基在肝癌组织较癌旁组织中显著上调表达,且其表达水平与病人病理分期呈正相关.用siRNA方法瞬时消减CK2β的表达,显著抑制肝癌细胞SMMC-7721的增殖及克隆形成率,并抑制细胞的迁移.消减CK2β能增强肝癌细胞对喜树碱等多种抗肿瘤药物的敏感性.这些发现提示CK2β亚基在肝癌的生长和迁移中发挥重要作用,可作为抑制肝癌细胞增殖的潜在药物靶点,为开发新的抗肿瘤药物提供线索. CK2, also known as casein kinase Ⅱ , is a kind of serine/threonine kinase which is highly conservative in eukaryotic cells. CK2 plays important roles in cell growth, proliferation, apoptosis and biological rhythm control. This paper investigated the role of the beta subunit of CK2 the in development and metastasis of Hepatocarcinoma. Firstly, the expression level of CK2β in HCC tissues and paired normal tissues was studied. Real-time PCR was used to compare the mRNA expression level of CK2β in 91 pairs of HCC samples. CK2 showed higher expression in 69 HCC tissue samples (75. 8% of the total samples) compared with the paired normal tissues. Immunohistochemistry staining of 75ps HCC tissue samples showed that CK219 expression was significantly higher in the tumor tissues. The expression level of CK2β in eytosome has a correlation with TNM clinicopathologie stage. Knocking down of CK2β by siRNA inhibits the growth, colonial formation and migration of SMMC-7721 ceils. Down-regulation of CK2β increases the sensitivity of cell to certain anti cancer drugs such as CPT. These new findings suggest that CK2β might be a key gene in cell growth and migration and a potential target of anti cancer drugs which provide clues for the development of new antitumor drugs.
