黄芩素联合左氧氟沙星对金黄色葡萄球菌生物膜的体外作用 被引量:13

Effect of baicalein combined with levofloxacin on Staphylococcus aureusbiofilm in vitro

摘要 目的探讨黄芩素协同左氧氟沙星对金黄色葡萄球菌生物膜的影响,为临床用药提供参考。方法构建生物膜体外模型;分为空白组、黄芩素组、阳性对照克拉霉素组、左氧氟沙星组、克拉霉素+左氧氟沙星组、黄芩素+左氧氟沙星组;两倍稀释法测定药物的最低抑菌及杀菌浓度;连续稀释法行活菌计数;激光扫描共聚焦显微镜及扫描电镜观察生物膜形态。结果 17 546株金黄色葡萄球菌经培养3d和7d后可形成早期及成熟期的生物被膜;早期活菌计数结果显示,无论是早期或成熟期各组单独作用时与空白组相比差异均无统计学意义,而早期的黄芩素+左氧氟沙星组(7.19±0.12)log10CFU/ml、克拉霉素+左氧氟沙星组(7.25±0.13)log10CFU/ml,与空白组(7.88±0.10)log10CFU/ml或左氧氟沙星(7.81±0.07)log10CFU/ml单独作用组相比,活菌计数明显减少(P<0.05);成熟期的黄芩素+左氧氟沙星组(7.29±0.09)log10CFU/ml、克拉霉素+左氧氟沙星组(7.36±0.11)log10CFU/ml与空白组(7.89±0.12)log10CFU/ml或左氧氟沙星(7.87±0.08)log10CFU/ml单独作用组相比,活菌计数也明显减少(P<0.05);激光扫描共聚焦显微镜观察发现无论是早期或成熟期药物组合作用组与单独作用组相比,死菌均明显增多。结论黄芩素对金黄色葡萄球菌生物膜具有破坏作用,与左氧氟沙星联合应用,可显著增强左氧氟沙星对金黄色葡萄球菌生物膜的杀菌作用。 OBJECTIVE To investigate the effect of baicalein combined with levofloxacin(LFX)on the Staphylococcus aureus biofilm(BF) so as to guide the clinical medication. METHODS The model of S. aureus biofilm was built in vitro; then the models were divided into six groups., the control group, baicalein group, clarithromycin(CLARY) group, LFX group, CLARY +LFX group,and the baicalein+LFX group; the MIC and MBC of test drugs were measured by the double dilution method; the bacterial colony counts in BF were recorded by serial dilution method; the shapes of S. aureus BF were observed through the confocal laser scanning microscope (CLSM). RESULTS A total of 17 546 strains of Early(3d) and mature(7d) S. aureus BF models were established successfully. The early bacterial colony count results showed that the difference in the bacterial colony count between the early S. aureus BF, mature S. aureus BF, and the control group was not statistically significant; the bacterial colony count was (7. 19±0.12)log10 CFU/ml in the early baicaleinq-LFX group, (7.25±0.13)log10 CFU/ml in the early CLARY+LFX group, as compared with (7. 88±0. 10)log10 CFU/ml of the control group and (7. 81 ±0.07)1og10 CFU/ml of the LFX group, the bacterial colony count decreased significantly (P〈0. 05); the bacterial colony count was (7.29±0.09)log10 CFU/ml in the mature Baicaleinff-LFX group, (7.36±0.11)log10 CFU/m in the mature CLARY+LFX group, as compared with (7.89±0.12)log10 CFU/ml of the control groupor (7. 87±0.08)log10 CFU/ml of the LFX group, the bacterialcolony count was also decreased significantly. The CLSM found that dead bacteria counts increased more significantly in either the early joint action group or the mature joint action group than did in the control group. CONCLUSION Baicalein can destroy the formed S. aureus BF and significantly reduce the bacterial colony count of S. aureus in BF when it is used in combination with LFX.
