IL-10和IL-33基因单核苷酸多态性与中山市汉族儿童哮喘易感性的关系 被引量:3

摘要 目的探讨IL-10和IL-33基因的单核苷酸多态性与中山市汉族儿童哮喘发病的相关性。方法采用病例对照研究,利用MassARRAY-IPLEX技术和基质辅助激光解吸电离飞行时间质谱平台(MALDI-TOF-MS)对158例儿童哮喘和198例健康对照的IL-10基因的rs1800896和IL-33基因的rs3939286位点进行基因分型。结果在中山市汉族儿童中rs1800896只检出AG和AA两种基因型,rs3939286检出AA、AG和GG 3种基因型,分型成功率为99.86%。与健康对照组相比,哮喘儿童rs1800896的基因型分布差异有统计学意义(2=3.864,P=0.049),但经Bonferroni校正后差异无统计学意义(Pc=0.098),等位基因分布差异无统计学意义(2=3.639,P=0.056)。rs3939286的基因型分布(2=9.988,P=0.007,Pc=0.014)和等位基因分布(2=8.266,P=0.004)差异有统计学意义。与GG基因型相比,rs3939286的AG和AA+AG基因型均能明显增加哮喘发生的危险性,OR值(95%可信区间)分别为2.230(1.243~3.997)和2.361(1.324~4.210)。与GG基因型相比,rs3939286的AA+AG基因型明显增加嗜酸性粒细胞计数(Z=-2.962,P=0.003)和IgE浓度(Z=-3.628,P=0.000)。结论 IL-33基因的rs3939286位点可能与中山市汉族儿童哮喘的发病有关。
