摘要
目的研究锌预处理对辣椒素致痛模型小鼠行为学变化的影响。方法 C57/BL6小鼠给予低锌或高锌预处理2 w后,利用足底注射辣椒素制备神经病理性疼痛(Neuropathic Pain,NPP)的动物模型。注射辣椒素后7 d,利用原子吸收光谱技术、热痛阈检测和机械痛阈检测技术观察小鼠脊髓锌离子的分布以及动物行为学变化。结果 小鼠足底注射辣椒素后,脊髓中的锌含量下降,动物热痛阈和机械痛阈降低(P<0.01);高锌预处理能显著增加脊髓中锌的含量,提高热痛阈和机械痛阈值;而低锌预处理组则加重脊髓的缺锌状况,使热痛阈和机械痛阈值更低(P<0.01)。结论 锌能提高NPP动物的痛阈,锌对NPP有抑制作用。
Objective To observe the influence of zinc pretreatment on the behavior of model micewith pain induced by capsaicin. Methods The C57/BL6 mice were pretreated with high or low zinc for 2 weeks firstly. Then capsaicin was injected into the sole to build mice model with neuropathic pain (NPP). After 7 days, atomic absorption spectroscopy, thermal and mechanical pain threshold detections were used to observe the distribution of the spinal cord zinc ions and ani- mal behavior change. Results The zinc level of spinal cord decreased in NPP model mice, and both the thermal and me- chanical pain threshold decreased ( P 〈 0. 01 ). Additionally, the high zinc pretreatment could increase the zinc level in spinal cord, and enhance thermal and mechanical pain threshold; while the low zinc pretreatment could aggravate the zinc deficiency in spinal cord, and decrease thermal and mechanical pain threshold ( P 〈 0. 01 ). Conclusion Zinc can in- crease the pain threshold of NPP model mice, and zinc can inhibit the NPP.
