摘要
目的 :建立非胰岛素依赖型糖尿病 ( NIDDM)模型 ,用于糖尿病研究。方法 :给予新生大鼠一次性腹腔注射链脲佐菌素 ,9周后检测糖耐量、血胰岛素水平等 ,并进行组织形态学检查。结果 :造模大鼠体重正常 ,饮水量和饮食量明显增多 ,空腹血糖、血胰岛素水平正常 ,但口服葡萄糖糖耐量低下、胰岛素分泌水平低下 ,第 1时相分泌明显减弱 ,并有延时分泌现象。病理学检查 ,造模大鼠胰岛体积明显缩小 ,β细胞减少。结论 :新生大鼠一次性腹腔注射链脲佐菌素可成功制备 NIDDM模型。
Objective:For preparation of non-insulin-dependent diabetes mellitus (NIDDM).Methods:Rats were intraperitoneally given 95mg/kg of streptozotocin (STZ) at one day of age. When rats were 9 weeks old, oral glucose tolerance test , plasma insulin determination, and morphological checks were performed.Results:In STZ-treated rats, the volume of water drinking and food intake was markedly increased; fast plasma glucose lever and insulin lever were normal, but oral glucose tolerance test and insulin secretion were lower; first-phase secretion of insulin decreased obviously, but a phenomena of delayed secretion was observed. In morphology, we observed that the area of islet and number of β-cell in pancreas reduced markedly.Conclusion:Giving neonatal SD rat STZ injection may make a stabilizing NIDDM model in rat.[
