摘要
目的 探索天然抗角蛋白自身抗体 (AK auto Ab)对培养角质形成细胞及鳞癌细胞凋亡相关蛋白的表达影响 .方法 经体外培养人角质形成细胞及鳞癌细胞 ,以不同滴度的天然抗角蛋白自身抗体作用 ,并用 MTT法及免疫组化检测 .结果 AK auto Ab对二者的增殖有着明显的抑制作用 ,且呈一定的剂量 -效应关系 .可促进二者凋亡相关蛋白 c- Myc的表达 ;对 Bcl- 2在 SCC中的表达表现为抑制作用 ,而对其在KC中的表达作用却不明显 .结论 天然抗角蛋白自身抗体对培养角质形成细胞及鳞癌细胞的增殖起着抑制作用 。
AIM To evaluate the mechanism that natural anti keratin autoantibody (AK auto Ab) inhibits the proliferation of keratinocytes (KC) and squamous cell carcinoma (SCC). METHODS Normal human KC and SCC were cultured and then treated with AK auto Ab in different titres. MTT and Immunohistochemical technology were used to test it. RESULTS AK autoAb inhibited the proliferation of KC and SCC with relation to dose effection. It also inhibited the expression of c Myc in KC and SCC. To Bcl 2, it had the same effect on SCC, but had little influence on its expression in KC, which was related to apoptosis. CONCLUSION AK auto Ab plays an important role in the proliferation of KC and SCC by regulating the protein related to apoptosis.
