经颅高压电烧伤大鼠可溶性细胞间黏附分子-1与白细胞黏附变化及乌司他丁干预作用 被引量:4

Changes of leukocyte adhesion and soluble intercellular adhesion molecule 1 expression induced by transcranial high-voltage electrical burns in rat and the therapeutic effects of Ulinastatin

摘要 目的了解经颅高压电烧伤对大鼠可溶性细胞间黏附分子-1(sICAM-1)的影响,探讨sICAM-1与白细胞黏附的关系及乌司他丁(UTI)干预效果。方法将180只清洁级SD大鼠按随机数字表法分为对照组、电伤组和治疗组,每组60只。电伤组和治疗组大鼠用调压器及实验变压器制成经颅高压电烧伤模型,对照组接相同装置但不通电致假伤。伤后2min内,对照组和电伤组大鼠经腹腔注射0.9%氯化钠溶液2mL,治疗组大鼠经腹腔注射2×104U/kgUTI注射液2mL。于伤前15min、伤后5min、1h、2h、4h、8h,每时相点采用随机数字表法于各组取10只大鼠,用布莱德多项投影显微镜系统检测肠系膜微静脉白细胞黏附数;经心脏采血用ELISA法检测血清sICAM-1含量。对数据行两因素析因设计的方差分析和LSD检验。结果 (1)电伤组sICAM-1含量高于对照组,差异有高度统计学意义(主效应F=536.550,P<0.01);电伤组伤后5min~8h各时相sICAM-1含量均高于本组伤前值,差异有高度统计学意义(主效应F=70.371,P<0.01)。治疗组sICAM-1含量低于电伤组,差异有高度统计学意义(主效应F=14.784,P<0.01);治疗组伤后5min~8h各时相sICAM-1含量均高于本组伤前值,差异有高度统计学意义(主效应F=126.391,P<0.01)。(2)电伤组白细胞黏附数多于对照组,差异有高度统计学意义(主效应F=1309.845,P<0.01);电伤组伤后5min~8h各时相白细胞黏附数均多于本组伤前值,差异有高度统计学意义(主效应F=73.485,P<0.01)。治疗组白细胞黏附数少于电伤组,差异有高度统计学意义(主效应F=447.666,P<0.01);治疗组伤后5min~8h各时相白细胞黏附数均高于本组伤前值,差异有高度统计学意义(主效应F=82.933,P<0.01)。结论经颅高压电烧伤引起大鼠血清sICAM-1含量升高,肠系膜微静脉白细胞黏附数量增加。UTI可降低经颅高压电烧伤大鼠血清sICAM-1含量,减少肠系膜微静脉白细胞黏附数。 Objective To investigate the influence of transcranial high-voltage electricity on leukocyte adhesion and soluble intercellular adhesion molecule 1 ( sICAM-1 ) expression in rats' blood serum and the therapeutic effects of Ulinastatin (UTI). Methods One hundred and eighty SD rats were randomly divided into three groups, control group, electrical burn group (burn group) and UTI-treated group (treatment group) and each group contained 60 rats. Voltage regulator and experimental transformer were used to make high-voltage electricity burns (HEB) animal model. The above equipment without electricity was used to treat the rats of the control group. 2 mL of normal saline (NS) were intraperitoneal injected into the rats of control or burn group. 2 mL of UTI (2 ×10^4 U/kg)were intraperitoneal injected into the rats of treatment group. Ten rats in each group were selected randomly at 15 minutes before HEB, 5 minutes later, 1 hour later, 2 hours later, 4 hours later and 8 hours later. Amount of adhesional leukocyte in the mesenteric venule was detected using the BVPM. sICAM-1 concentration in the serum of the cardiac blood was assayed with ELISA. And the experimental data were analyzed statistically with variance analysis and LSD test. Results ( 1 ) slCAM-1 expression in burn group was increased compared with that in control group and the change was significant statistically (F = 536. 550,P 〈 0.01 ). sICAM-1 expression at 5 minutes to 8 hours after the burn was higher than that before the burn and the increase was significant statistically ( F = 70. 371, P 〈 0. 01 ). sICAM-1 expression in treatment group was reduced compared with that in burn group and the reduction was significant statistically (F = 14. 784,P 〈0.01 ). sICAM-1 expression of treatment group at 5 minutes to 8 hours after the burn was higher than that before the burn, and the change was significant statistically (F = 126. 391 ,P 〈 0.01). (2) Amount of adhesional leukocyte was increased compared with that in control group, and the increase was significant statistically ( F = 1309. 845, P 〈 0.01 ). The amount in burn group at 5 minutes to 8 hours after the burn was higher than that before the burn and the change was significant statistically ( F = 73.485,P 〈0.01 ). Amount of adhesional leukocyte in treatment group was decreased compared with that in burn group and the reduction was significant statistically (F =447. 666,P 〈 0.01 ). The amount in treatment group at 5 minutes to 8 hours after the burn was higher than that before the burn and change was significant statistically (F = 82. 933, P 〈 0. 01 ). Conclusions Transcranial HEB up-regulated slCAM-1 expression in blood serum of rats and amount of adhesional leukocyte in the mesenteric venule. And UTI reduced the sICAM-1 expression and the amount of adhesional leukocyte after transcranial HEB.
