摘要
目的通过正交试验优选确定忍冬藤中有效成分绿原酸和咖啡酸水煎提取工艺条件。方法以加水量、提取次数和提取时间为考察因素,每个因数分别选取三个水平进行试验设计,以高效液相色谱测定绿原酸、咖啡酸提取含量为考察指标进行L9(34)正交试验。以直观分析法和方差分析法分析研究水提忍冬藤中绿原酸、咖啡酸的影响因素,确定最佳提取工艺。结果按提取忍冬藤中绿原酸、咖啡酸含量的影响因素大小顺序,依次为提取液用量、提取次数和提取时间。最佳提取工艺为A3B2C3组合,即15倍水煎煮3次,每次时间分别为30 min、60 min和30 min。结论正交试验优选新工艺条件稳定,快速简便,重现性较好,对忍冬藤中绿原酸和咖啡酸提取含量可提高近15%,为生产工艺改进提供指导。
Objective To select the optimal extracting procedure for chlorogenic acid and caffeic cid from Lonicera Japonica Thunb. Methods The experiment design was made using orthogonat from L9(34), the extraction times, aquavolume, and extraction time were the three factors. Each of the three factoxs was tested at three levels. Results The experiment results showed that the factors of the aquavolume and extraction times were more significant than the extraction time factor in the extracting procedure. The op.timal extracting procedure was extracting third times by using fifteen times of aqua, the first is 30 min, then 60 min, and the 30 min. Conclusion The factor of the aquavolume in the experiment of orthogonal design was the most important factor in the extracting procedure, and this extraction procedure is helpful for further study of extractiving technology of gallic acid of herba euphorbiae humifusae.
