基因重组人血管生长素衍生物Asp116His对缺血心肌再血管化的影响

摘要 在建立家兔心肌梗死模型的基础上 ,以基因重组人血管生长素衍生物Asp1 1 6His于缺血心肌边缘部作心肌多点注射。术后 8~ 1 0天取注射点心肌切片作HE染色、VB染色及Ⅷ因子标记的免疫组化染色观察 ;术后 1 8~ 2 0天 ,做非选择性冠脉造影。病理学检查 ,可见心肌间质内大量小血管结构 ,毛细血管明显增多 ,冠脉造影 ,示有侧支血管形成。本研究首次证实血管生长素衍生物Asp1 1 6His在心肌作用局部可引起明显新生血管形成 ,提示在心脏冠状血管内存在其有效作用位点。 In order to establish its angiogenic potential in ischemic myocardium, recombinant human angiogenin derivative Asp116His was applied in an acute myocardial infarction model in rabbits by direct intramyocardial injection into the border zone of the ischemic myocardium. Some of the animals were sacrificed on the 8th~10th postoperative day, With their hearts harvested and subjected to histologic studies. The rest animals underwent non-selective coronary angiography on the 18 th ~20 th postoperative day. Myocardium around the injection sites was apparently less ischemic in the experiment group as compared with control groups. A large number of small vessels were observed in the interstitial tissues and even more within the myocardium on immunohistochemistry studies. Angiographic studies showed formation of collateral vessels in the treated animals. The results indicate for the first time that angiogenin derivative Asp116His may act on ischemic myocardium and stimulate neo-angiogenesis, which holds great promise for its future application in biologic revascularization of ischemic myocardium.
