人血管生长素衍生物的重组腺病毒制备 被引量:4

摘要 目的 :构建并鉴定血管生长素 (ANG)衍生物重组腺病毒 ,为心肌缺血区促血管生长因子基因转染研究作准备。方法 :ANG衍生物 D116 H - ANG重组粘粒与腺病毒 DNA末端肽复合物混合后以磷酸钙共沉淀法转染 2 93人胚肾细胞株细胞。结果 :酶切结果显示 ,重组粘粒中 ,D116 H - ANG插入方向正确 ,所获重组腺病毒带有 ANG衍生物基因。结论 :所获重组腺病毒为 E1 ,E3缺陷型 。 Objective:To construct and confirm the D116H ANG recombinant adenovirus for the studies on gene transfection of the angiogenic factors to the ischemic cardiac muscle. Methods: D116H ANG pAxCAwt cosmid DNA and DNA TPC were mixed and co transfected to the 293 cell by calcium phosphate coprecipitation. Results: The result of restriction showed the direction of the insertion was right and the recombinant adenovirus generated contained the gene of D116H ANG. Conclusion: The recombinant adenovirus generated is E 1 and E 3 deleted which can be further used in gene therapy in vivo .
