利用基因芯片筛查主动脉夹层外周血白细胞差异基因 被引量:1

摘要 目的主动脉夹层(aortic dissection,AD)发病后血液内环境发生的病理改变会造成全身性损伤,外周血白细胞基因表达谱的改变是病理改变的基础。文中筛选AD外周血白细胞差异表达基因。方法提取AD和主动脉瘤患者外周血白细胞总RNA,以RNA为模板逆转录获取cDNA,用生物素标记cRNA,纯化后用Agilent基因芯片扫描分析基因表达谱的变化,获得差异表达基因。结果筛选出515个差异表达基因,其中上调基因238个,下调基因277个,功能未知者占40%。功能已知者主要涉及炎症反应、免疫反应、细胞骨架和胶原调节、氧化应激、代谢、凝血、细胞凋亡等相关基因。结论 AD发生伴有白细胞差异基因表达,应用基因芯片筛查差异基因,对阐明AD发病过程中病理生理机制和寻找标志基因有重要意义。 Objective The pathological changes of blood internal environment can cause systemic damage in aortic dissec- tion, the gene expression profile of peripheral blood leukocyte is the key factor, but there were few studies at present. The present stud- y was to screen the differentially expressed genes of peripheral blood white blood cells in aortic dissection. Methods RNA was ex- tracted from peripheral blood white blood cells of aortic dissection and aortic aneurysm patients, reversed transcripted to cDNA, then marked by biotin, purified and hybrided with Agilent gene chip, finally, the chip was scanned and analysed for differentially expressed genes. Results Five hundred and fifteen differentially expressed genes were detected, including 238 up-regulated and 277 down- regulated known genes, and 40% unknown function gene. The differentially expressed genes mainly involves the inflammatory reaction and immune response, collagen protein adjustment, oxidative Stress, metabolism, coagulation , cell apoptosis and other biological processes. Conclusion .The differentially expressed genes screened from white blood cells in aortic dissection is an important mean to elucidate the mechanic pathophysiology change in aortic dissection and get biomarks genes by gene chip.
