摘要
目的 了解辛德毕斯病毒基因结构与功能的关系 ,阐明其分子致病分子机理。方法 构建了辛德毕斯病毒 (XJ- 16 0病毒株 )结构基因的真核表达质粒 ,pcDNA3.1ABC ,转染BHK - 2 1细胞后观察其对宿主细胞的影响。结果 转染病毒结构基因的BHK - 2 1细胞 ,可以发生辛德毕斯病毒感染所出现的细胞毒性反应 ,表现为 :细胞存活率降低 ;细胞核染色质浓缩、凝集 ;G1期细胞减少、而处于S期的细胞增多。
Objective To study the effect of Sindbis XJ-160 virus structural proteins on BHK-21 cell. Methods We constructed the expressing vector pcDNA3.1ABC that contains the XJ-160 virus structural protein gene,and then transfected it to BHK-21 cell.Results The expressed products can be detected by RNA dot blot hybridization or immnofluorescent assay.The cytopathic effect was observed in BHK-21 cells transfected with pc DNA3.1ABC. There was also condensed chromatin found accumulated.BHK-21 cell death and marked cell cycle changes were observed. Conclusions These results indicate that cell appoptosis might be one of the mechanisms inducing cell death by XJ-160 virus and its structural protein gene probably plays a role in this programmed cell death.
