摘要
目的:建立保健食品中金丝桃苷测定的高效液相色谱(HPLC)方法。方法:试验采用迪马钻石Ⅱ(4.6 mm×250 mm,5μm)色谱柱,乙腈-0.2%磷酸水溶液为流动相洗脱,在波长为256 nm下进行检测。结果:方法的线性范围:9.37 g/ml~187 g/ml。相关系数r为0.9999;检出限:0.4μg/g;定量限:1.2μg/g。高低两个浓度水平加标回收率:87%~94%;相对标准偏差(RSD)1.8%。结论:本方法简便、快速、准确、重现性好,适用于保健食品中金丝桃苷的测定。
Objective:To develop a method for determination of Hyperin in health food by high performance liquid chromatography(HPLC).Methods: The target analytes were separated on Diamonsil C18(Ⅱ)(4.6 mm×250 mm,5 μm)column with acetonitrile-0.2% H3PO4 as mobile phase for gradient elution and detected at the wavelength of 256 nm.Results: The calibration curves of Hyperin showed good linearity in the range of 9.37 g/ml^187 g/ml.The correlation coefficient was 0.9999.The detection limit was 0.4 μg/g.The quantitative detection limits of the method was 1.2 μg/g.The average recoveries at two concentration levels were 87%~94% and the relative standard deviation(RSD) of the method was less than 1.8%.Conclusion: The method is simple,rapid,accurate and reproducible,it can be used for the determination of Hyperin in health food.
