摘要
采用 mRNA差异展示技术分离正常和经(60)COγ射线照射的人肠上皮细胞株 HIEC细胞之间的差异表达基因,为进一步揭开电离辐射对胃肠细胞损伤的分子机理奠定基础。从正常的和经γ射线照射的人肠上皮细胞中分离出差异条带共101条,31个为新表达的差异片段,29个为高表达的差异片段, 41个为要表达的差异片段;其中 10个片段属于 D-T(11)G组, 59个片段属于 D-T(11)A组; 32个片段属于 D-T(11)C组。从新表达的差异片段中随机挑取 5个片段作探针,与正常的及照射后的人肠上皮细胞总RNA进行打点杂交,进一步证实呈差异表达。结果表明,101个差异表达基因与γ辐射损伤有关,其中31个新表达基因可能与电离辐射引起正常组织损伤关系更密切。
In order to isolate differentially expressed genes of intestinal epithelium cell related with r radiation, by using HIEC cell line in vitro culture as research target, the cell line was irradiated by 8Gy of r ray and was continued to culture for 24h as radiation group, tile cell line without r radiation was as control group. We isolated the differentially expressed genes between radiation group and control group by mRNA differential display. We obtained 101 fragments of differentially expressed genes, 31 of which were new expressed genes from radiation group, 29 of which were overexpressed genes which was higher expressed in radiation group than in control group, 41 of which were no expression in radiation group and expression in controlgroup, 31 of which were from the group of D-T_11G as anchored primer, 59 of which were from the group of D-T_11A as anchored primer, 32 of which were from the group of D-T_11C as anchored primer. In summary, we obtained 101 fragments of differentially expressed genes related with r radiation, 31 among them may be closely correlated with radiation damage to intestinal.
