摘要
目的检测基质金属蛋白酶7(MMP-7)及碱性成纤维细胞生长因子(bFGF)在口腔鳞状细胞癌(OSCC)中的表达,并探讨其在OSCC发生发展中的作用。方法应用逆转录-聚合酶链反应(RT-PCR)技术对36例手术切除的OSCC组织及14例正常口腔组织(NOT)中MMP-7及bFGF mRNA的表达进行检测。结果 MMP-7及bFGF mRNA在OSCC表达均高于NOT(P<0.01),在有淋巴结转移组的表达均高于无淋巴结转移组(P<0.01),两者的表达均与肿瘤的病理分期、淋巴结转移有关,MMP-7及bFGF表达之间呈正相关(r=0.621,P<0.01)。结论 MMP-7和bFGF的异常表达与OSCC生物学行为密切相关,成为预测OSCC侵袭与转移的标志物,并为患者的预后提供评估依据。
AbsObjective To detect the expression of MMP-7 and bFGF in oral squamous cell carcinoma(OSCC) and to explore the role they play in progress of OSCC. Methods The expression of MMP-7 and bFGF mRNA in 36 cases of OSCC and 14 cases of normal oral tissue were detected by reverse RT-PCR. Results The positive expression rates of MMP-7 and bFGF mRNA in OSCC were significantly higher than those in normal oral tissue (P 〈 0.01 ). Compared with those without lymph node metastasis in OSCC, the positive ratios of MMP-7 and bFGF mRNA with lymph node metastasis were significantly higher( P 〈 0.01 ). The expressions of MMP-7 and bFGF were significantly related with pathological stage and the lymph node metastasis,but neither of them was correlated with sex and age. Pearson rank correlation analysis showed that bFGF expression level was positively related to MMP-7 level (r = 0. 621, P 〈 0.01 ). Conclusion The abnormal expressions of MMP-7 and bFGF are closely related to the biological behavior of OSCC, so they may be used as markers in predicting the invasion and metastasis of OSCC and provide evaluation criterion for the prognosis.
