氮红霉素的合成工艺研究

摘要 以红霉素E肟(Ⅰ)为原料,经贝克曼重排反应得到6,9-亚胺醚(Ⅲ)后,再经NaBH4/7H2O.CeCl3催化体系还原及水解等步骤最终得到高收率、高纯度的氮红霉素(Ⅴ)。以化合物Ⅰ计算,化合物Ⅴ的收率为91.7%,经HPLC检测纯度为99.2%。此次研究改变了重排反应和还原反应中的催化剂,使整个工艺的收率大大提高,且成本有所下降。 Azaerithromycin is obtained with high yield and high purity by the reduction and hydrolysis of erythromycin 6, 9-imine ether,which was prepared by the beckmann rearrangement reaction of erythromycin E oxime. The yield is 86.9% calculated as the weight of raw material, and the purity is 98.4%. This study changes the catalysts in rearrangement reaction and reduction, which greatly improves the yield and reduces the cost of the whole process.
