摘要 目的:探讨蕲艾鞣酸对人体肿瘤的治疗作用。方法:采用超声提取方法对蕲艾鞣酸进行提取,通过3-(4,5-二甲基噻唑-2)-2,5二苯基四氮唑溴盐法(MTT法)在体外检测不同浓度的蕲艾鞣酸对人肝癌细胞HepG2的细胞毒性作用。在此基础上通过大孔树脂柱层析对蕲艾总鞣酸正丁醇部位提取分离,得到5个不同极性的鞣酸馏分,并通过MTT法比较不同极性的各提取物对肝癌细胞的抑制活性。结果:蕲艾鞣酸使体外培养的人肝癌细胞HepG2的增殖效率下降,且呈浓度与时间的依赖关系,具有一定的细胞毒性。结论:5个不同极性的提取物均具有抑制肝癌细胞生长的作用,其中40%甲醇部位对肝癌细胞的生长抑制作用最强,是蕲艾鞣酸体外抗肝癌肿瘤主要的活性部位。 Objective:To investigate the role of Chinese mugwort tannic acidon human tumor. Methods:The tannic acid were extracted by ultrasonic of the Chinese mugwort, using MTT to determine the cytotoxicity of different concentrations of Chinese mugwort tannic acid on human hepotoma cells HepG2. On this basis, n - butyl alcohol of Chinese mugwort tannic acid was extracted and separated by column chromatography of macroporous resin, had five different polarity fractions of tannic acid in Chinese mugwort, anti -tumor activity of the above extraction was determined by MTT assay. Results: Chinese mugwort tannic acid inhibited proliferation of human hepatoma HepG2 cells line,in a dose and time- dependent manner,with certain cytotoxic. Conclusion:Five different polar extractions had the anti - tumor activity,in which the 40% methanol inhibited cell proliferation of liver cancer line is the most strong one, the 40% methanol from Chinese mugwort tannic acid had anti -tumor activity in vitro.
