液质联用法测定辣椒红色素中罗丹明B含量 被引量:11

Determination of Rhodamine B in Capsicum Red Pigment by High Performance liquid Chromatography Coupled with Tandem Mass Spectrometry

摘要 建立了食品添加剂辣椒红色素中违禁物质罗丹明B的超高效液相色谱-串联质谱测定方法。样品经80%甲醇水(含2%甲酸)溶液提取后,甲醇饱和正己烷净化,以乙腈和0.3%甲酸水为流动相进行梯度洗脱,采用BEH C18柱分离,正离子模式检测,在多反应监测模式下进行定性定量分析。罗丹明B在0.50μg/L~102.20μg/L浓度范围内线性关系良好,相关系数R2>0.999,相对标准偏差RSD<3.4%,方法回收率为86.7%~96.7%,定量限为0.5μg/kg。该方法处理简单、灵敏度高、分析时间短,适合分析辣椒红色素中罗丹明B含量。 A method based on high performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS)was developed for determination of rhodamine B in capsicum red pigment. The sample was ex- tracted with 80 % methanol-water (containing 2 % formic acid)and then cleaned up by methanol saturated hex- ane. The target analytes was separated on BEH Cls column with gradient elution us acetonitrile and water (con- taining 0.3 % formic acid)as mobile phases. Under the optimized detection conditions ,the linear range for rho- damine B was 0.50μg/L-102.20 μg/L,The limits of quantification was 0.5 μg/kg . The relative standard devia- tions of reproducibility less than 3.4 %. The recoveries of the method were between 86.7 % and 96.7 %. The method is simple, rapid, highly sensitive and suitable for determination rhodamine B in capsicum red pigment.
