摘要
采用一种新离子交换介质Express IonC(简称IonC)使鼠神经生长因子 (mNGF)能得到规模纯化。以鼠颌下腺为原料 ,匀浆后 ,经Express IonC纯化得单一组分的mNGF ,用SDS PAGE测定其相对分子质量约为 13.7× 10 3,比活约 2 .3× 10 5U mg。结果表明IonC可替代CM 5 2 ,且更适于神经生长因子的规模化生产。
Using a new ion?exchange chromatography to purify mNGF on large scale. The purification procedure include tissue homogenization and twice Express?IonC Chromotography. The molecular weight(MW) was measured by means of SDS?PAGE. The bioactivity was assayed by the method of dorsal root ganglion. A homogenous product of MW is 13.7 kD. The activity of the product was about 2.3×10 5 U/mg. The results indicated that Express?IonC may instead of CM?52 and Express?IonC is best suit for large scale purification.
