响应面分析法优化葡萄皮色素提取工艺 被引量:6

Optimization of extraction technique for pigment from grape skin with response surface methodology

摘要 以葡萄皮为研究对象,以吸光度为衡量提取工艺的指标。在单因素试验的基础上,选取提取时间、料液比、提取剂乙醇浓度为自变量,吸光度为响应值,利用Box-Behnken中心组合设计原理和响应面分析法,研究各变量及其交互作用对吸光度的影响,模拟得到二次多项式回归方程的预测模型,在pH值为1、温度为70℃的条件下,确定最佳提取条件为:乙醇浓度51%,料液比1∶12,提取时间117 min。在此工艺条件下,吸光度为0.682,与理论预测值0.7097相比,其相对误差约为3.90%,说明通过响应面优化后得出的方程具有一定的实践指导意义。 With grape skin as the material and the absorbance as the measure indicator, three extraction parameters including extraction time, solid-liquid ratio and extractant(ethanol) concentration were optimized using central composite design and response surface methodology based on single factor investigations for achieving maximum the absorbance. The interaction of the respective variables and their influence on the absorbance were studied by using Box-Benhnken central composite design and response surface analysis theory, the simulated quadratic polynomial regression equation of prediction model was set up. Under the condition of pH 1 and extraction temperature 70℃, the optimum extraction condition was determined as: solid-liquid ratio 1:12, ethanol concentration 51%, extraction time of 117 min. Under these conditions, the average absorbance was 0.682 and compared to the theoretical value 0.7097, the relative error of 3.90%.
