摘要
以发根农杆菌 A4 菌株介导 ,对宁夏枸杞叶片和茎切段的遗传转化进行了初步研究 ,建立了发状根体系 ,并优化了转化条件。乙酰丁香酮的添加、农杆菌液的浓度、共培养时间、外植体取材部位及时间 ,均可以影响发状根的诱导频率。采用添加 1 0 0 μmol/L的乙酰丁香酮、振荡培养 2 4 h的 A4 菌液感染 3周龄的叶片切段 ,并共培养 3d,可以得到最佳的转化效果 ,发状根的诱导率为 48.9%。在同样的转化条件下 ,只有 1 3.6%的茎切段上能诱导出发状根。在感染菌液之前 ,外植体预培养 3d对发状根的诱导也有一定的促进作用。
Genetic transformation of leaf and stem segments from Lycium barbarum L.mediated with Agrobacterium rhizogenes A 4 strain was investigated,and hairy root lines and optimal condition of transformation were obtained.The addition of acetosyringone,the concentration of bacterium suspension,the time of inoculation,different explants as well as seedling age exhibited certain influences on transformation.The optimal transformation of leaf segments was achieved by the use of Agrobacterium A 4 strain cultivated for 24 h in the YMB medium supplemented with 100 μmol/L acetosyringone,and leaf segments from 3 week seedlings and by a 3 day coculture.The induction frequency was 48.9%,where as the frequency of stem segments was only 13.6% on the same condition.A 3 day preculture of recipient explants before bacterium infection premoted the formation of hairy root.
