大豆胰蛋白酶抑制剂酶法钝化及应用研究 被引量:1

Enzymatic deactivation of soybean trypsin inhibitor and the application in soybean protein bioprocessing

摘要 分析了不同大豆原料和产品中胰蛋白酶抑制剂的含量,研究了大豆胰蛋白酶抑制剂对猪胰蛋白酶的抑制作用、抑制剂的酶法钝化及钝化技术在大豆蛋白酶解中应用。大豆胰蛋白酶抑制剂达到纯猪胰蛋白酶40%以上的含量时能完全抑制胰蛋白酶活性,达到工业胰蛋白酶3%以上的含量时可抑制最高70%的蛋白酶活性。碱性蛋白酶能有效酶解钝化大豆胰蛋白酶抑制剂,用量为大豆胰蛋白酶抑制剂含量的25%,作用时间5h。0.6%碱性蛋白酶+0.6%胰蛋白酶组合酶解10%豆浆,水解度比2种蛋白酶单独酶解时明显提高,达到24%。酶法钝化大豆胰蛋白酶抑制剂,对大豆食品的深加工和营养性能的提高具有重要价值。 The content of trypsin inhibitor in a variety of soy ingrdients and products was analyzed,and the inhibition rate of trypsin inhibitor,the deactivation technology and its application in soybean bioprocessing were also studied.The proteinase activity of pure porcine trypsin was completely inhibited by ratio of 40% soybean trypsin inhibitor,and up to 70% proteinase activity of industrial trypsin was inhibited by more than ratio of 3% soybean trypsin inhibitor.Alcalase effectively deactivated soybean trypsin inhibitor with the optimal enzyme/substrate(E/S) ratio of 0.25 at 5 h.Alcalase 0.6%+trypsin 0.6% hydrolyzed 10% soymilk and the hydrolization degree reached up to 24% which was apparently higher than that for single alcalase or trypsin hydrolyzation.The result might be valuable for soybean protein bioprocessing and its nutritional improvement.
