摘要
目的:以紫花羊耳蒜无菌带芽块茎为外植体,建立其高频再生体系。方法:采用不同培养基、不同配比植物激素及不同移栽基质等,对紫花羊耳蒜进行丛生芽诱导、壮苗生根及炼苗移栽研究。结果:芽增殖的最适合培养基为1/2MS(大量元素减半)+6-BA 3.5 mg/L+NAA 0.2 mg/L,增殖系数平均达到3.24倍;适合紫花羊耳蒜离体培养壮苗生根的培养基配方为1/2MS+6-BA 0.6 mg/L+NAA 0.5 mg/L+20%的马铃薯汁,生根率可达95%;紫花羊耳蒜组培苗长出3条根时可炼苗,移栽基质以菜园土∶兰花土=1∶1为宜,移栽成活率为63.64%。结论:以紫花羊耳蒜无菌带芽块茎为外植体,能够实现其高频再生体系建立,为其人工扩大栽培奠定了良好的基础。
Objective: To establish the rapid propagation system of Liparis nigra Seidenf. by the aseptic tubers with buds. Methods: Different media, plant growth regulators at different concentration and different material were used to L.nigra Seidenf. for cluster buds induction and multiplication, rooting and transplanting. Results: The medi- um 1/2MS+6-BA 3.5 mg/L+NAA 0.2 mg/L was most suitable for buds induction and multiplication, where the mul- tiplication was 3.24. The optimal medium for strengthing seedlings and rooting seedlings was 1/2MS+6-BA 0.6 rag/ L+NAA 0.5 mg/L+20% potato juice sucrose. The rooting rate was up to 95%. The seedlings could be used for transplanting from training bottles to the greenhouse when had 3 roots. The garden soil:orchid soil=1: 1 was the best material where the survival rate could be up to 63.64%. Conclusion: The rapid propagation system of can be established by the aseptic tubers with buds, which could promote the artificial cultivation of L.nigra Seidenf.
