摘要
目的探讨乙酰葛根素对Aβ_(1-42)海马立体定位注射导致阿尔茨海默病模型大鼠的药物作用及对海马小胶质细胞和炎症因子PKC-δ表达水平的影响。方法将40只Wistar大鼠随机分为空白对照组、模型组、乙酰葛根素低、高剂量组。应用Aβ_(1-42)双侧海马注射制备AD大鼠模型,Morris水迷宫测定不同时期各组大鼠的学习记忆能力,电镜观察小胶质细胞的活化程度,Elisa测定血清PKC-δ含量的变化。结果乙酰葛根素能显著改善阿尔茨海默病模型大鼠的学习记忆能力,减轻小胶质细胞的活化程度,降低PKC-δ的表达(P<0.05)。结论乙酰葛根素对阿尔茨海默病具有显著的预防和保护作用。
Objective To investigate the effect of acetyl puerarin on the hippoeampus stereotactic injection of Aβ1-42 model rats and tile expression of microglia and inflammation factors of PKC-δ. Methods Forty wistar rats were randomly divided into the control group, AD model group,aeetyl puerarin low-dose group and high-dose group. AD model rats was produced by bilateral hippocampal injection of Aβ1-42. The rats'different time of learning and memory was tested through the method of Morris water maze and electron microscope to investigale the activation of microglia,Elisa determination to detect the changes of PKC-δ. Results Acetyl puerarin significantly improved the learning and memoryability in AD rats and reduced the activation of microglia and the expression of PKC-δ ( P 〈 0.05 ). Conclusion Acetylpuerarin has obvious preventive and protective effect on Alzheimer disease.
