清瘟败毒饮对大鼠急性肺损伤保护作用的研究 被引量:8

STUDY ON PROTECTIVE EFFECT OF QINGWEN BAIDU LIQUID ON RATS’ ACUTE LUNG INJURY

摘要 目的:观察清瘟败毒饮对急性肺损伤(acute lung injury,ALI)大鼠肺组织病理学变化的影响。方法:实验大鼠随机分为正常对照组,模型组,泼尼松组和清瘟败毒饮大、中、小剂量组,每组24只。采用脂多糖气管内滴注诱导ALI模型,造模后连续用药3天。于末次给药后24、48小时每组各处死6只大鼠,72小时处死余下大鼠,并称取大鼠肺湿重和干重,计算肺干湿比和肺系数;收集肺泡灌洗液行白细胞计数;取右肺中叶行HE染色,光镜观察肺组织病理变化。结果:清瘟败毒饮各剂量组大鼠BALF中白细胞总数均降低,肺泡损伤、炎症细胞浸润和红细胞渗出情况有较大程度的改善,病理学炎症积分(除72小时小剂量组外)均降低,清瘟败毒饮大剂量组大鼠肺干湿比及各剂量组大鼠肺系数均下降,与同时相模型组相比均有显著性差异或非常显著性差异(P<0.05或P<0.01)。结论:清瘟败毒饮对脂多糖致大鼠急性肺损伤有保护作用。 Objective: To observe the effect of qingwen baidu liquid on pathological changes of lung tissue in rats with acute lung injury induced by lipopolysaccharide. Methods: Rats were divided into six groups,normal,model,predrison,low,middle,high dose qingwen baidu liquid groups,each group contained 24 rats. Models with acute lung injury were made by lipopolysaccharide intratracheal instillation,each group was given the corresponding medicine or physiological saline for three days,twice a day,six rats from each group were killed at 24 and 48 hours after giving medicine at the last time,the other rats were all killed at 72 hours. Than taking rats’ lungs to weigh wet weight and dry weight,counting the lung coefficient and ratio of wet and dry weight,collecting bronchoalveolar lavage fluid(BLF) to count the number of white blood cells,taking the middle lobe of rats’ right lung to observe the pathological changes of lung tissue in rats by light microscope. Results: To compare with model group,the number of white blood cells in bronchoalveolar lavage fluid of rats in qingwen baidu liquid groups was obviously decreased,alveolar injury,inflammatory infiltration and red blood cells exudation were improved in the three dose groups,pathological inflammatory intergral was significantly reduced in the three qingwen baidu liquid groups,the ratio of wet and dry lung weight in qingwen baidu liquid high dose group and lung coefficient in the three dose groups were decreased obviously,there were significant differences between the three dose liquid groups and model group on above aspects. Conclusion: Qingwen baidu liquid can protect acute lung injury of rats induced by lipopolysaccharide.
