摘要
目的观察痛泻要方对溃疡性结肠炎(UC)大鼠结肠组织病理形态及血清中相关因子的影响,探讨其作用机制。方法 60只SPF级Wistar大鼠随机分为空白组、模型组、柳氮磺胺吡啶(SASP)组及痛泻要方低、中、高剂量组,除空白组外,其余各组大鼠均以2,4,6-三硝基苯磺酸(TNBS)/乙醇灌肠法制备UC大鼠模型,同时,痛泻要方低、中、高剂量组分别按原药材11、22、44 g/kg剂量灌胃,SASP组按0.3 g/kg剂量灌胃,灌胃体积均为10 mL/kg,空白组、模型组灌服等体积生理盐水,持续3周。HE染色观察大鼠结肠组织病理形态并评分,采用ELISA法检测大鼠血清白细胞介素(IL)-1β、肿瘤坏死因子(TNF-α)和IL-4水平。结果 HE染色显示,各治疗组结肠组织病理形态明显改善;与空白组比较,模型组大鼠血清IL-1β、TNF-α含量均明显升高,IL-4含量显著下降(P<0.05,P<0.01);与模型组比较,痛泻要方高、中、低剂量组大鼠血清IL-1β、TNF-α水平降低,而IL-4水平明显升高(P<0.05,P<0.01)。结论痛泻要方通过下调促炎因子IL-lβ、TNF-α的表达,升高抗炎因子IL-4的表达,调节肠道异常免疫反应、抑制炎症反应,修复溃疡,进而防治UC。
Objective To observe the curative effect and mechanism of Tongxieyao Formula on ulcerative colitis(UC).Methods Sixty SPF Wistar rats were randomly divided into blank group,model group,SASP group and low-,medium-,high-dose of Tongxieyao Formula groups.Except the blank group,UC models were made by means of TNBS/ethanol enema.The low-,medium-,high-dose of Tongxieyao Formula groups were given Tongxieyao Formula 11,22,44 g/kg by gavage,SASP group was given 0.3 g/kg,the volume was 10 mL/kg.The blank group and model group were given with the same volume of normal physiological saline for three weeks.The pathological morphology of colon was observed and scored by hematoxylin-eosin staining(HE),IL-1β,TNF-α and IL-4 in rat serum were detected by ELISA.Results Compared with the model group,the colon tissue pathological morphology was significantly improved in all treatment groups.Compared with the blank group,the content of IL-1β and TNF-α in the model group was increased,and the content of IL-4 was decreased significantly(P 0.05,P 0.01).The content of IL-1β and TNF-α in high-,medium-,low-dose of Tongxieyao Formula groups were lower than that in the model group,and the content of IL-4 was higher(P 0.05,P 0.01).Conclusion Tongxieyao Formula can decrease the expression of lL-lβ and TNF-α,increase the expression of IL-4,regulate immune abnormalities of intestine,inhibit inflammation,repair ulcers,and then prevent and treat UC.
