摘要
[目的]利用活体生物发光成像技术,探讨中国被毛孢菌丝体对小鼠乳腺癌肺转移的影响。[方法]4T1-luc乳腺癌细胞原位接种于50只BALB/c小鼠2周后,取40只荷瘤小鼠,分成4组,即模型组(10mL.kg-1NS)、HSM低剂量组(1.0g.kg-1HSM)、HSM高剂量组(2.0g.kg-1HSM)、阳性组(20mg.kg-1CTX),每组10只,连续给药21d;测定各组小鼠原位肿瘤体积和重量,并用活体生物发光成像仪观察各组小鼠乳腺癌肺转移情况。[结果]小鼠灌胃给予1.0g.kg-1、2.0g.kg-1HSM后,原位肿瘤体积和重量明显降低(P<0.05,P<0.01),原位肿瘤抑瘤率分别为22.92%和32.50%;给予2.0g.kg-1HSM的小鼠乳腺癌肺转移明显减少(P<0.01),乳腺癌肺转移的抑制率为51.77%。[结论]中国被毛孢菌丝体具有较好的抑制小鼠乳腺癌肺转移作用。
[Objective] To investigate the effects of inhibiting breast cancer lung metastasis of Hirsutella sinensis mycelium by bioluminescent imaging(BLI).[Method] Fifty BALB/c mice were injected 4T1-luc breast cancer cells in situ.After two weeks,forty tumor-bearing mice were selected to divide into four groups,model group,low-dose group(1.0g·kg-1 HSM),high-dose group(2.0g·kg-1 HSM) and positive group(20mg·kg-1 CTX),ten mice for each group.After 21 days of administration,observing lung metastasis from breast camcer by BLI in vivo,and determing tumor volume and weight of mice in situ.[Result] Situ tumor volume and weight were significantly reduced after treatment with 1.0-2.0g·kg-1 HSM in mice,with the inhibition ratio of 22.92% and 32.50% respectively.The lung metastasis from breast cancer was significantly reduced after treatment with 2.0g·kg-1 HSM in mice,with the inhibition ratio of 51.77%.[Conclusion] Hirsutella sinensis mycelium has excellent inhibition of lung metastasis and breast cancer in mice.
