摘要
目的 :观察缺氧对培养心肌细胞DNA单链的损伤情况 ,以探讨缺氧对心肌细胞的损伤机制。方法 :实验分为对照组、缺氧 3小时组、缺氧 6小时组、缺氧 12小时组和缺氧 2 4小时组 ,采用单细胞凝胶电泳技术检测心肌细胞DNA的单链损伤。结果 :缺氧 6小时后 ,心肌细胞核DNA发生了单链断裂 ,损伤细胞的百分率和断裂损伤程度均明显增加 ,与对照组差异有显著意义 (P <0 0 5 ) ;随缺氧时间的延长 ,DNA的损伤程度和受损伤心肌细胞的百分率均增加 ,与对照组差异有极显著意义 (P <0 0 1)。结论 :缺氧能够导致心肌细胞DNA的单链断裂损伤 ,其中DNA的单链断裂损伤明显早于双链的断裂损伤 。
Objective:To observe the damages of DNA single strand induced by hypoxia in cultured neonatal rat cardiomyocytes.Methods:Cultured neonatal rat cardiomyocytes were randomly assigned to control group,hypoxia for 3h group,hypoxia for 6h group,hypoxia for 12h group and hypoxia for 24h group.The DNA single strand damages were detected with Single Cell Gel Electrophoresis in individual cardiomyocyts.Results:In comparison with control group,the DNA single strands were damaged significantly after hypoxia for 6h and more seriously with hypoxia for a longer time( P< 0 01).Conclusion:Hypoxia can induce DNA single strand damages in the cultured cardiomyocytes,which might occur more early than DNA double strands damages do.It suggests that the DNA single strand damages might be one of important mechanisms of cardiomyocytes apoptosis induced by hypoxia.
