摘要
目的 :探讨流式细胞仪 (FCM)在膀胱癌多药耐受研究中的应用。方法 :利用蛋白质印迹、免疫细胞化学和 FCM检测并比较膀胱癌多药耐受细胞 EJ/MRP的多药耐受糖蛋白 (P- gp)和多药耐受相关蛋白 (MRP)的表达 ;利用 FCM检测了 EJ/MRP药物耐受的功能水平。结果 :EJ/MRP高表达 MRP,无明显 P- gp表达 ,FCM的检测结果与蛋白质印迹、免疫细胞化学的结果一致 ,但检测更简便 ;功能检测显示柔红霉素 (DNR)可以作为 FCM检测 MRP功能的发光探针 ,细胞外排药物的功能增强至少是 MRP高表达的膀胱癌细胞内药物聚集减少的一个重要原因。结论 :FCM是检测膀胱癌多药耐受蛋白质表达水平和 MRP功能水平的好方法 ,DNR是 FCM检测MRP功能的良好探针。
Purpose:To observe the application of flow cytometer (FCM) in multidrug resistant (MDR) study of bladder carcinoma.Methods:The expression of P-glycoprotein (P-gp) and multidrug resistant associated protein (MRP) of EJ/MRP,a multidrug resistant cell line of bladder carcinoma,was tested by western blot,immunocytochemistry and FCM method.The function level of drug resistance of EJ/MRP was also detected by FCM.Results:EJ/MRP over-expressed MRP but without obvious P-gp expression.The results of protein detection by FCM were consistent with those by western blot and immunocytochemistry,but more convenient.The function detection showed daunorubicin (DNR) could be used as a probe to detect the function of MRP by FCM,and the increase of efflux of intracellular drug was at least an important cause to lead to the decrease of intracellular drug accumulation.Conclusions:FCM is a good method to detect MDR protein and MRP function of bladder carcinoma,and DNR is a good probe to detect MRP function by FCM.
