甜菜TR1位点多态性分析及细胞质育性鉴定 被引量:5

TR1 allele polymorphism analysis and cytoplasm fertility identification for 8 sugar beet material populations

摘要 本研究针对8个甜菜材料的细胞质育性进行分子鉴定。利用数目可变串联重复序列(Variable Number of Tandem Repeats,VNTR)分子标记技术,分别对8个甜菜材料的线粒体DNA中TR1位点VNTR片段多态性进行分析。分析显示8个甜菜群体线粒体TR1位点VNTR重复序列分别为6拷贝,4拷贝,13拷贝。标记分析结果表明田间选育的8个甜菜群体的细胞质分别属于不育型,保持型,普通育性恢复型三种细胞质类型。并且发现1个保持系细胞质群体中混有恢复系细胞质单株和细胞质不育型单株,1个不育系群体中混有普通授粉系单株,1个正常花粉材料群体为不育型细胞质。该分子标记鉴定结果为这些材料的育性进一步决选提供了分子依据,淘汰田间选育过程中混杂的单株,弥补田间表型育性鉴定结果的不足。 Molecular identification for cytoplasm fertility type of 8 sugar beet materials was conducted in this paper. VNTR polymorphisms in mtDNA TR1 allele for sugar beet populations were analyzed by Variable number of tandem repeats (VNTR) molecular markers. The result shows that the Numbers of Tandem Repeats in TR1 locus for 8 sugar beet populations are six, four and thirteen copy. Molecular markers analysis results indicate that the cytoplasm DNA of 8 sugar beet populations breeding from field belong to male sterility, maintain ( o - type), and normal fertility restore type. One o -type population mixed with normal fertility restore and male sterility plants. One normal fertility restore type plant was detected in one male sterility population. One pollen viable population belongs to male sterility cytoplasm DNA. The study provides molecular basis for materials key selecting and weed out mix single plant. The result can also make up the shortage of field phenotype fertility identification.
