共轭亚油酸对奶牛乳腺上皮细胞脂肪酸摄取与转运相关基因mRNA转录的影响 被引量:2

Effect of conjugated linoleic acid on transcription of mRNAs of genes related to the uptake and transport of fatty acid in dairy cow mammary-gland epithelial cells

摘要 为了研究共轭亚油酸(CLA)对奶牛乳腺上皮细胞脂肪酸摄取与转运相关基因mRNA转录的影响,试验采用组织块法体外培养奶牛乳腺上皮细胞,外源添加不同浓度的c9,t11-CLA与t10,c12-CLA作用乳腺上皮细胞48 h,应用实时荧光定量PCR技术,测定相关基因mRNA的转录。结果表明:1)与对照组相比,两种CLA均显著降低了脂蛋白脂肪酶(LPL)基因mRNA的表达丰度(P<0.05);2)150μmol/L c9,t11-CLA显著上调分化抗原簇36(CD36)基因mRNA的转录水平(P<0.05),其转录水平随t10,c12-CLA浓度增加显著上升(P<0.05);3)150μmol/L c9,t11-CLA和不同浓度的t10,c12-CLA均可显著上调乙酰辅酶A结合蛋白(ACBP)的转录(P<0.05);4)与对照组相比,两种CLA均显著下调脂肪酸结合蛋白3(FABP3)基因mRNA的转录水平(P<0.05)。说明两种CLA均显著下调LPL mRNA和FABP3 mRNA的转录水平,150μmol/L的两种CLA异构体上调CD36和ACBP的转录水平。 To study the effect of conjugated linoleic acid (CLA) on transcription of mRNAs of genes related to the uptake and transport of fatty acid in dairy cow mammary - gland epithelial ceils, dairy cow mammary - gland epithelial ceils were cultured in vitro by tissue blocks. The dairy cow mammary - gland epithelial cells were treated with different concentrations of exogenous c9, t11 - CLA and tl0, c12 - CLA for 48 h, and then the transcription of mRNAs of related genes was determined using the real - time fluorescence - based quantitative PCR technology. The results showed that the two CLA isomers both significantly decreased LPL mRNA abundance, compared with the control group ( P 〈 0.05 ) ; 2 ) 150 μmol/L e9, t11 -CLA significantly upregulated the transcription level of CD36 (P 〈 0. 05 ), and its CD36 mRNA transcription level was significantly increased with the increased concentrations of t10, c12 - CLA (P 〈 0.05 ) ; 3) 150 μmol/L c9, t11 - CLA and different concen- trations of t10, c12 - CLA significantly upregulated the transcription of acyl - CoA - binding protein (ACBP) gene ( P 〈 0.05 ) ; 4 ) the two CLA isomers both significantly downregulated the transcription of fatty acid - binding protein 3 ( FABP3 ), compared with the control group ( P 〈 0.05 ). The results indicate that the two CLA isomers both significantly downregulate the mRNA transcription levels of LPL and FABP3, and two CLA isomers ( 150 μmol/L) can upregulate the transcription levels of CD36 and ACBP.
