生菜rbcS基因启动子序列的克隆与分析 被引量:4

摘要 根据其他植物中已知的光合作用关键酶1,5-二磷酸核酮糖羧化/加氧酶小亚基编码基因(rbcS)及其启动子序列设计简并引物,以生菜叶片基因组DNA为模板,利用PCR扩增技术获得生菜rbcS上游1 046 bp的序列元件(GenBank登录号:JQ741945)。序列分析表明,该序列元件包含植物启动子所必需的核心元件CAAT-box和TATA-box,同时包含具有参与光应答、茉莉酸应答、脱落酸应答、乙烯应答、热胁迫应答及分生组织激活等相关的多种调控元件。序列比对表明,该序列与菊科、茄科、豆科3种科属多种植物的rbcS启动子序列存在较高同源性。该启动子的获得为开展以生菜作为外源蛋白表达宿主的相关研究奠定了良好基础。 The 1 046 bp 5′ upstream regions of ribulose-1,5-bisphosphate carboxylase/oxygenase small subunit gene(rbcS)was amplified from lettuce genome DNA by PCR under the direction of the primers,which were designed based on the rbcS gene and the promoter sequence of other plants.Sequencing analysis indicated that the fragment contains core elements CAAT-box and TATA-box,as well as the elments sensitive to light,MeJA,abscisic acid,ethylene,and heat stress.Meristem expression regulatory element was also found in the cloned rbcS promoter sequence.Furthermore,the rbcS promoter sequence of lettuce is similar to the rbcS promoters isolated from other plants of Asteraceae,Solanaceae or Fabaceae.This work provides strong support for high-level expression of recombinant proteins in lettuce.
