摘要
肿瘤的发生不仅与细胞的增殖和异常分化有关,也和细胞凋亡异常有关。Survivin是凋亡抑制蛋白家族(inhibi-tor of apoptosis protein,IAP)的重要成员,它抑制细胞凋亡并在细胞分化中起重要作用。它在分化正常的细胞或组织中较少表达,但在肿瘤中大量表达;该基因可促使肿瘤组织生长,提高肿瘤组织的化疗耐药性并降低患者生存率,阻断其表达可以促进细胞凋亡并抑制肿瘤生长。因此,Survivin已作为肿瘤治疗的一种新靶点而被广泛研究。文中对Survivin在胰腺癌中的研究进展情况作一综述。
The development of tumor is not only related to cell proliferation and abnormal differentiation but also to abnormal apoptosis. Survivin is a structurally unique member of the inhibitor of apoptosis protein (IAP) family that acts as a suppressor of apop- tosis and plays a central role in cell division. Owing to its barely detectable nature in the terminally differentiated normal cells/tissues but massive upregulation in human tumors and its association with chemotherapy resistance, increased tumor recurrence, and shorter patient survival, disruption of survivin induction pathway can increase the apoptosis and inhibit tumor growth, thus survivin is currently under extensive investigation as a novel therapeutic target. In this text, the new progress of survivin study in pancreatic carcinoma is re- viewed.
