氧化型染发剂主要成分对大鼠皮肤角朊细胞DNA损伤作用的研究 被引量:10

摘要 应用大鼠皮肤角朊细胞的原代培养技术并结合单细胞凝胶电泳法检测氧化型染发剂的两种主要成分双氧水(H2 O2 )、对苯二胺 (PPD)及二者混合物致大鼠皮肤角朊细胞DNA的单链断裂作用。结果表明 ,染发剂及其组分均能使大鼠角朊细胞DNA发生单链断裂 ,染毒剂量与慧尾长度呈剂量 反应关系 (rH2 O2 =0 978,rPPD=0 993,rH2 O2 -PPD=0 996 ) 。 The single cell gel electrophoresis assay was applied to detect the occurrence of DNA single strand breaks in rat keratinocytes after exposed to the two main components of oxidation hair dyes p phenylenediamine(PPD)and hydrogen peroxide(H\-2O\-2)and their mixture.The results showed that exposure of rat keratinocytes to the samples could cause DNA single strand breaks of the cells.Dose responsive relatonships between exposure and the extent of DNA strand breaks (r\-\{H\-2O\-3\}=0\^978,r\-\{PPD\}=0\^993,r\-\{H\-2O\-2\}=0\^996) were found.These results showed that the keratinocyte cytotoxicity of oxidation hair dyes might be considered as a result form its capacity to cause DNA strand breaks,and they also explored the genotoxicity mechanism of oxidatoin hair dyes.
