摘要 目的 :研究雷公藤多甙和IL 10对DC内IL 12p40和DC CK1转录的影响。方法 :通过GM CSF、IL 4和TNFα体外培养体系 ,从人外周血单个核细胞中诱导DC ,IL 12p40和DC衍生的趋化因子 1(DC CK1)转录采用逆转录多聚酶链反应 (RT PCR)技术。结果 :通过GM CSF、IL 4和TNFα体外培养体系可以获得成熟功能性DC ,雷公藤多甙和IL 10能抑制DC内IL 12p40mRNA的转录 ,但对DC CK1mRNA的转录无明显影响。结论 :雷公藤多甙、IL 10可能通过抑制IL 12p40mR Objective: To investigate the influences of tripterygium wilfordii (TⅡ) and interleukin 10 (IL 10) on interleukin 12 (IL 12) p40 subunit and dendritic cell derived C C chemokine 1 (DC CK1) transcription of human dendritic cells (DC). Methods: DC were generated by culturing peripheral blood mononuclear cells of healthy volunteers with GM CSF、IL 4 and TNFα in vitro. Reverse transcriptase PCR analysis was used to detect IL 12 p40 and DC CK1 mRNA transcription. Results: TⅡ(20μg/ml) and IL 10 (200ng/ml) inhibit the transcription of IL 12 p40, but there is no significant effect on the transcription of DC CK1. Conclusion: TⅡ and IL 10 interfered with the function of human DC by inhibiting transcriptions of IL 12 p40 subunit.
