摘要 目的:研究雷公藤多苷多次给药的免疫抑制作用及安全范围。方法:用迟发性变态反应,淋巴细胞转化和血清溶血素测定方法,观察雷公藤多苷免疫抑制作用;小鼠连续灌胃给药7 d,观察急性毒性反应。用bliss法分别计算各药效指标坪值的ED_(50),ED_(95),ED_(99)和LD_(50),LD_5,LD_1,计算雷公藤多苷治疗指数(TI),安全系数(SF)和可靠安全系数(CSF)。结果:雷公藤多苷有明显的免疫抑制作用,呈良好的量效关系,相关系数r分别为0.982,0.940,0.972,其安全范围的均值TI,SF和CSF分别为5.9,1.5和0.9。结论:雷公藤多苷有显著的免疫抑制作用,但安全范围窄,小剂量多次给药相对安全。 AIM:To research the margin of safety and suppressing immunity of Tripterygium wilfordii polyglycosidum (TWP) with multiple administration. METHODS: Applying the methods of delayed hypersensitivity reaction,the lymphocyte transfer and serum hemolysin test observed the immunosuppression effect of TwP. Mice were fed with TwP. The acute toxic reaction for 7 d and the median effective doses (ED50) ,ED95,ED99, median lethal dose (LDso) ,LD5 and LDI of plateau value were calculated by bliss method. The therapeutic index (TI), safety factor(SF) and certain safety factor (CSF) of TwP were assured by applying-some formulas. RESULTS: The immunosuppression effect of TwP was obviously detected with good quantity-efficiency relationship. The correlation coefficients(r) were 0.982,0.940,0.972. TI,SF and CSF safety scopes were 5.9,1.5 and 0.9 respectively. CONCLUSION: TWP can significantly suppress immunity,but the safety scope is narrow. It would be more safe with administration of splitting small doses.
