CD4^+CD25^+FOXP3^+Treg与脑血管支架植入后的炎症相关性研究 被引量:1

The inflammatory response of CD4^+CD25^+FOXP3^+Treg cells in patients underwent cerebral stent implantation

摘要 目的探讨颅内及颈部支架植入患者外周血中CD4+CD25+FOXP3+调节性T细胞(Treg细胞)随访3个月的变化及意义。方法入选40例行颅内及颈部血管支架植入患者(根据NIHSS评分分为2组)进行支架植入术前、术后7 d以及术后3个月进行随访,同时与正常人群(20例)进行对照。采用流式细胞术、Real-Time PCR和ELISA分别对外周血中Treg细胞的频率、转录调节因子FOXP3 mRNA水平和相关细胞因子TGF-β的表达进行检测。结果 NIHSS>2分患者外周血中CD4+CD25+FOXP3+Treg/CD4+T细胞比例和FOXP3的mRNA水平以及TGF-β在术后7 d较术前和正常组是显著降低的(P<0.05),在术后3个月显著上升,且与正常组无显著性差异(P>0.05),NIHSS≤2分患者Treg在随访过程中无显著性变化。结论 Treg细胞可能参与了金属裸支架植入后引发的局部和系统的急性炎症反应,且后期增加的Treg细胞与抑制术后慢性炎症反应有关。 We aimed to investigate the alteration of Treg in patients underwent intracranial or cervical stents implantation and its significance in the 3 months follow-up time. Of the total 40 enrolled patients underwent stent implantation, 21 patients with NIHSS score ≤2 and 19 patients with NIHSS score 〉2 were divided into two groups for subsequent investigations and 20 age-matched volunteers were enrolled as controls. The frequencies of the CD4+ CD25+FOXP3+Treg cells, the mRNA level of FOXP3, and the expression of TGF-β in peripheral blood were detected by flow cytometric analysis, real-Time PCR, and ELISA, respectively. Results showed that there was no difference of Treg level in patients with NIHSS ≤2 seores before and after stent implantation as compared with the control. In patients with the NIHSS〉2 scores, Treg level was decreased at 7^th day and returned to normal levels in 3rd month. We speculate that the maintenance of the frequency and function of Treg is involved in controlling the inflammation response, which otherwise causes the acute inflammation after stent implantation
