摘要
骆驼科动物体内存在一种只含重链不含轻链的天然重链抗体,克隆骆驼体内重链抗体的可变区后,得到的仅由一个重链可变区组成的单域抗体,称为纳米抗体。纳米抗体分子质量小,具有稳定性强、可溶性好、易表达、免疫原性弱、穿透性强、靶向性强、人源化简单等特点,使其在肿瘤、感染性疾病、肠炎、淀粉样变疾病、血栓以及动脉粥样硬化病变的诊断和治疗方面具有良好的应用效果,同时可以显著降低成本。基于骆驼重链抗体的VHH单域抗体的特殊结构,其在基础研究、新药的开发、疾病的诊断和治疗等方面具有广阔的前景。
Functional heavy-chain antibodies lacking light chains occur naturally in Camelidae animals. The single domain antibodyies are constructed by cloning the variable domains of heavy chain antibodies, and refferred to nanobodies. Owing to the properties like smaller size, high stability, good solubility and targe- ting effect, easy expression, low immunogenicity, great penetration, simple humanization, nanobody has great application effect in the diagnosis and therapy of tumor, infectious diseases, enteritis, amyloidosis, thrombosis and atherosclerotic lesion, meanwhile, it also reduce costs. Based on the special structure of single domain antibody that derived from heavy chain antibody, it has great application prospects in drug development, basis research and disease diagnosis and therapy.
