摘要
目的:分析细胞凋亡抑制蛋白survivin在原发性肝癌(HCC)及其癌旁组织中的表达,探讨其在肿瘤血管形成和抵抗肿瘤血管内皮细胞凋亡中的作用。方法:采集HCC癌组织、癌旁组织和正常肝组织标本,通过免疫组织化学方法分析survivin蛋白表达阳性率与HCC病理参数的关系,采用流式细胞术分析survivin蛋白在不同组织中的表达水平,用荧光定量PCR方法分析不同组织中survivin基因的表达水平。结果:survivin蛋白在肝癌组织中表达水平与门静脉癌栓的形成有关联(P<0.05),与肿瘤大小、包膜、甲胎蛋白(AFP)水平及HCC病理分期均无关联(P>0.05);流式细胞术结果,肝癌组织中survivin蛋白表达水平高于癌旁组织和正常肝组织(P<0.05),癌旁组织中survivin蛋白表达水平高于正常肝组织,但差异无统计学意义(P>0.05);荧光定量PCR检测,survivin基因只在肝癌组织中表达,在癌旁组织和正常肝组织中无表达,其表达水平组间比较差异有统计学意义(P<0.05)。结论:细胞凋亡抑制蛋白survivin在肝癌组织中表达水平较高,但在癌旁组织中检测不到其基因的表达,survivin蛋白可能是通过旁分泌作用从肝癌病变处进入癌旁组织细胞中发挥抑制细胞凋亡的功能。
Objective To analyze the survivin expressions in primary hepatocellular carcinoma (HCC) tissue and para-carcinoma tissue, and to investigate the effects of survivin on tumor blood vessel growth and inhibiting apoptosis of tumor endothelial cells. Methods The human HCC, para-carcinoma and normal liver tissues were collected, and the relationship between the positive rate of survivin protein expression and pathological parameters of HCC was analyzed by immunohistoehemitry, and the survivin protein expression levels in different tissues were detected by flow cytometry, and the survivin gene expression levels in different tissues were measured by quantitative real-time PCR (qRT-PCR). Results The survivin protein expression in HCC tissue was related to portal vein tumor thrombus formation (P〈0.05), and had no relationship with tumor size, casple, AFP level and pathological stages of HCC (P〈0.05). The flow cytometry results showed that the survivin protein expression in HCC tissue was higher than those in para-carcinoma and normal liver tissues (P〈0.05); the survivin protein expression in para-carcinoma tissue was higher than that in normal liver tissues, but there was no significant difference (P〈0.05). The qRT-PCR results showed that the survivin mRNA was only detected in HCC tissue, not detectable in para-earcinoma or normal liver tissues. Conclusion Survivin is highly expressed in HCC tissue, hut the survivin gene is not expressed in para-carcinoma tissue, and the survivin protein may transport through a paraerine manner from HCC.
