纤维素酶法提取杏鲍菇多糖工艺优化 被引量:26

Opitimization of extraction processing on polysaccharides from Pleurotus eryngii by cellulase enzymolysis

摘要 利用纤维素酶处理杏鲍菇提取多糖,在单因素试验基础上选取试验因素与水平,根据中心组合试验设计原理采用3因素3水平的响应面分析法,对提取工艺进行优化设计,建立了多糖得率测算模型的回归方程,确认酶加量、酶解时间和酶解温度对提取多糖得率都有显著影响。在分析各因素的显著性和交互作用后,得出纤维素酶提取杏鲍菇多糖的最佳工艺条件:料液比1:30、酶加量0.35%、酶解温度50℃、酶解时间2h、酶提1次,纤维素酶提取杏鲍菇多糖的提取率高达18.57%,结果表明酶法比传统水提取方法多糖提取率高1.24倍。 On the base of single-factor experiments and the principles of Box-Behnken central composite expermmental design, response surface methodology (RSM) with three factors and three levels was used to optimize the extraction processing of polysaccharides from Pleurotus eryngii treated with cellulase and establish the quadratic regression equation for the yield of polysaccharides .The results showed that the enzyme amount, the enzymolysis time and the enzymolysis temperature have significant effects on the yield of polysaccharides, and the optimal extraction prosessing conditions of polysaccharides were as follows: ratio of material solvent of 1:30, enzyme amount of 0.35%, enzymolysis temperature of 50 ℃, enzymolysis time of 2 h and extraction of 1. Under the conditions above, the extraction rate of polysaccharides from Pleurotus eryngii was up to 18.57%. The comparison of extraction efficiency between the methods treated with and without cellulose indicated that the extraction efficiency of total polysaccharides could be improved by 1.24 times th^ough the treatment with cellulase.
