UPLC simultaneous determination of five components in Naoxintong capsules

摘要 目的:建立超高效液相色谱(UPLC)同时测定脑心通胶囊中羟基红花黄色素A、芍药苷、阿魏酸、丹酚酸B、藁本内酯5个主要化学成分的方法。方法:采用WATERS ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7μm),流动相为0.5%甲酸-乙腈梯度洗脱,流速为0.3 mL·min-1,检测波长400 nm(羟基红花黄色素A)、235 nm(芍药苷),280 nm(丹酚酸B),324nm(阿魏酸和藁本内酯)。结果:本方法可在24 min内完成一次色谱分析,各主要成分色谱峰之间有良好的分离度,羟基红花黄色素A、芍药苷、阿魏酸、丹酚酸B、藁本内酯5个成分质量浓度分别在0.970~9.70 mg·L-1(r=0.9993)、3.38~33.8 mg·L-1(r=0.9995)、0.336~3.36 mg·L-1(r=0.9996)、2.15~21.5 mg·L-1(r=0.9999)、1.46~14.6 mg·L-1(r=0.9998)的范围内与峰面积呈良好的线性关系,方法的平均回收率(n=6)分别为99.47%、101.3%、98.45%、98.54%、99.02%,精密度、重复性的RSD均小于2.0%。结论:本方法能同时测定脑心通胶囊中5个主要化学成分,可较全面地检测脑心通胶囊的质量。 Objective: To establish a UPLC method for simultaneous determination of five compounds( hydroxysaf- flor yellow A, paeoniflorin, ferulic acid, salvianolic acid B, ligustilide) in Naoxintong capsules. Methods: The five compounds in Naoxintong capsules were simultaneously determined by UPLC with WATERS ACQUITY UPLC BEH C18 column(2. 1 mm × 100 mm,1.7 μm). The mobile phase consisted of 0.5% formic acid aqueous -acetonitrile with a gradient elution program and the flow rate of 0. 3 mL ~ rain - ~, the determination wavelengths were 400 nm for hydroxysafflor yellow A,235 nm for paeoniflorin,280 nm for salvianolie acid B ,and 324 nm for ferulic acid and li- gustilide. Results: The five components were completely separated in 24 min at one time. There were good separa- tion degrees between each chromatic peak. The linearity was achieved in the range of 0. 970 - 9.70 mg · L- 1 ( r = 0. 9993) ,3.38 -33.8 mg · L-1(r =0. 9995) ,0. 336 -3.36 mg · L-1 (r =0. 9996) ,2. 15 -21.5 mg · L-1(r- 0. 9999 ) , 1.46 - 14.6 mg ·L - 1 ( r = 0. 9998 ) for hydroxysafflor yellow A, paeoniflorin, ferulic acid, salvianolic acid B, ligustilide, respectively. The average recoveries ( n = 6 ) were 99.47 %, 101.3 %, 98.45 %, 98.54% , 99.02 % , re- spectively. The RSDs of precision and reproducibility were all less than 2.0%. Conclusion: The method can simul-taneously determine the five major chemical compounds in Naoxintong capsules, and can be used for overall quality control of Naoxintong capsules.
