摘要
研究了东北野猪耳皮组织成纤维细胞的分离、培养、纯化方法和生长特性,并对培养细胞进行了污染检测及冷冻保存和复苏培养观察。结果显示:原代培养在接种5d组织块边缘开始游离出单个细胞,到10d细胞生长旺盛;用酶消化法和差速贴壁法得到纯化的东北野猪成纤维细胞;冷冻前和复苏后的细胞活率分别为96.7%和92.7%,差异不显著(P>0.05);分离纯化的东北野猪成纤维细胞生长曲线正常;支原体和病毒检测呈阴性;成功建立东北野猪成纤维细胞系。
The isolation and culture, purification methods and growth characteristics of northeast- ern wild boar ear skin fibroblasts were studied, and the fibroblasts contamination were detected and the cryopreservation of cultured cells and recovery were observed. The results showed that the northeastern wild boar fibroblasts were isolated by attaching tissue explants from ear skin,in- oculated with 5 days the tissue edge to free single cells,inoculated with 10 days fibroblast growth vigorous. The fibroblasts were purified by enzyme digestion and differential adhesion method. The survival rate of fibroblasts before cryopreservation and after resuscitation were 96. 7% and 92.7%,no significant difference(P〉0. 05). The northeastern wild boar ear skin fibroblasts growth curves were normal. Tests for virus and mycoplasma were negative. The northeastern wild boar ear skin fibroblasts line was successfully established.
