摘要
目的建立成纤维细胞系及细胞库。方法采用体外细胞培养技术进行皮肤成纤维细胞的分离、培养、传代、冻存、复苏及鉴定。结果人皮肤成纤维细胞传代至23代以上,此时的细胞数是原代培养的细胞数的数十万倍,经冻存、复苏后,人皮肤成纤维细胞仍具有增殖能力,生长状态与原代培养的人皮肤成纤维细胞相类似。结论酶消化法是培养人皮肤成纤维细胞简便、快捷的方法。
Objective To establish human skin fibroblasts line and bank.Methods Cell culture in vitro technique was used, including isolation,culture,subculture,frozen storage,recovery and identification of human fibroblasts, in this study.Results The human fibroblasts were passed at least 23 passages with more than 223 times the original plating number.After frozen storage and recovery,the fibroblasts could proliferate and differentiate,which showed similar growth conditions as primarily cultured fibroblasts.Conclusion The fibroblasts bank has been established.This technique (i.e enzyme-digective method) for establishment of human fibroblasts bank is easy, simple and feasible under routine laboratory conditions.
