摘要
局部黏着斑激酶(focal adhesion kinase,FAK)是一种非受体型酪氨酸蛋白激酶,是重要的细胞内骨架蛋白,并参与多种信号通路。研究发现,FAK在许多肿瘤中过表达,通过多种途径参与肿瘤的发生、发展过程,且与恶性肿瘤的浸润转移有关,尤其与食管癌、肝癌和结肠癌等消化系肿瘤关系密切。FAK已经被当作潜在的肿瘤治疗靶点来研究。因此,研究FAK与消化系肿瘤的关系在肿瘤防治中具有重要意义。本文结合国内外文献对FAK在消化系肿瘤发生发展及其转移过程的研究进展作一综述。
Focal adhesion kinase (FAK) , a nonreceptor tyrosine kinase, which is a intra-cellular skeleton protein and takes part in many kinds of signal pathway. It showed that FAK over expressed in many kinds of tumor and partici- pated in the genesis and development of various tumors in many ways. In addition, FAK is associated with the invasion, metastasis of malignant tumors, especially gastrointestinal tumors such as esophageal cancer, liver cancer, colon cancer, etc. FAK has already taken as potential cancer treatment targets to research. So it is significant to study the relationship between FAK and gastrointestinal tumors for tumor prevention and treatment. In combination with domestic and overseas literatures, this article provides a brief review of the research progress in the roles of FAK in the genesis, development and metastasis of gastrointestinal tumors.
