摘要
目的:建立灵芝及保健食品中三萜类化合物的含量测定方法。方法:样品经乙酸乙酯提取,三萜类化合物在高氯酸和香草醛的热酸性条件下可反应产生有色物质成分,利用其产生特征性的颜色,依据有色成分颜色的不同吸收度计算含量,其显色灵敏而稳定,比色样品溶液在548nm波长处测定吸收。结果:对照品熊果酸的检测浓度在2.0μg/ml~24.0μg/ml范围内与吸光度呈良好的线性关系,r=0.9991,平均回收率97.49%,RSD为2.21%。结论:分光光度法实用,灵敏、快速、相对准确、成本低。
Objective:To establish a method for determination of triterpenoids in ganoderma and health food. Methods: Triterpenoids were extracted using acetic ether and produced substances with a characteristic color under hot acid conditions with the presence of perehloric acid and vanillin; the coloration was sensitive and stable. The c.ontent of triterpenoids was calculated based on the absorbance of the sample solution measured at 548 nm. Results: A good linear relationship was demonstrated between the absorbance values and the concentrations of the reference substance, ursolic acid, over the range of 2.0 μg/ml - 24.0 μg/ml ( r = 0. 9991 ) ; the mean recovery rate was 97.49% and the RSD was 2.21%. Conclusion: Spectrophotometry is practical, sensitive, rapid and relatively ac- curate and cost - effective ; it is applicable to determination of triterpenoids in ganoderma and health food.
