摘要
目的:探讨胃癌组织中类泛素特异性蛋白酶1(sentrin-specific protease 1,SENP1)的表达与临床病理特征间的关系,及其与血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平、微血管密度(microvessel density,MVD)间的关系。方法:采用免疫组化染色EnVision法,以SENP1和VEGF抗体检测胃癌及胃良性病变(胃炎、胃溃疡)组织中SENP1、VEGF蛋白的表达;用EnVision法以CD34抗体标记血管内皮细胞,并计数MVD。结果:SENP1蛋白在胃癌组织中的阳性率为75.0%(75/100),而在胃良性病变组织中的阳性率为36.0%(18/50),差异有统计学意义(P<0.01);SENP1的表达与胃癌的淋巴结转移、浸润深度、肿瘤分化程度及临床分期等临床指标间有相关性(P<0.05)。VEGF蛋白在胃癌组织中阳性率为71.0%(71/100),而胃良性病变中其阳性率为12.0%(6/50),差异有统计学意义(P<0.01);VEGF的表达与胃癌的淋巴结转移、浸润深度及临床分期等临床指标间有相关性(P<0.05)。胃癌组织中MVD为32.93±14.05,而胃良性病变中其为28.67±8.17,差异有统计学意义(P<0.05);MVD与胃癌的淋巴结转移、浸润深度及临床分期等临床指标间有相关性(P<0.05)。SENP1蛋白与VEGF蛋白的表达呈正相关(r=0.219,P<0.05),VEGF蛋白表达与MVD呈正相关(r=0.199,P<0.05),SENP1蛋白表达亦与MVD呈正相关(r=0.237,P<0.05)。结论:SENP1在胃癌组织中存在过度表达,参与了胃癌的发展,其可能通过调控缺氧诱导因子1调节VEGF的表达,在胃癌血管生成中发挥重要作用。SENP1、VEGF的过度表达及高MVD与胃癌的发生、发展有关,可能成为预测胃癌侵袭、转移及预后的重要指标。
Objective To investigate the expressions of sentrin-specific protease I (SENP1) and vascular endothelial growth factor (VEGF)and mierovessel density (MVD) in gastric cancer, and the relation of these factors with gastric cancer. Methods EnVision immunohistochemical staining method was used to detect the expressions of SENP1 protein and VEGF protein in gastric cancer and benign gastric lesion (gastritis and gastric ulcer). CD34 antibody was labeled to blood vessel endothelial cells and MVD was counted by EnVision method. Results The positive rate of SENP1 protein in gastric cancer (75.0%, 75/100) was significantly higher than that in benign gastric lesion (36.0%, 18/50) (P〈0.01). The expression of SENP1 was correlated with the degree of infiltration and differentiation, lymph node metastasis and TNM stage (P〈0.05). The positive rate of VEGF protein in gastric cancer (71.0%, 71/100) was significantly higher than that in benign gastric lesion (12.0%, 6/ 50) (P〈0.01), and the expression of the VEGF was correlated with the degree of infiltration, lymph node metastasis and TNM stage (P〈0.05). MVD in gastric cancer (32.93+14.05) was significantly higher than that in benign gastric lesion (28.67-+8.17) (P〈0.05), and MVD was correlated with the degree of infiltration, lymph node metastasis and TNM stage (P〈0.05). Positive correlations were found between expressions of SENP1 and VEGF (r--0.219, P〈O.05), expression of VEGF and MVD (r= 0.199, P〈0.05), and expression of SENPland MVD (r=-0.237, P〈0.05). Conclusions Development and progression of gastriccancer was associated with overexpressions of SENP1 and VEGF and high MVD. These biomarkers could be used to predict metastasis and prognosis, SENP1 has the potential to be used as a new target for gastric cancer treatment.
