弓形虫DNA疫苗免疫鼠体内重组质粒分布的探讨 被引量:4

STUDY ON DISTRIBUTION OF RECOMBINANT PLASMID IN MICE IMMUNIZED BY DNA VACCINE OF TOXOPLASMA GONDII

摘要 目的 通过肌注弓形虫 DNA疫苗质粒 p BK- P30、p BK- P2 2直接免疫小鼠 ,观察重组质粒在鼠体内不同器官组织的分布。方法 疫苗 p BK- P30、p BK- P2 2经肌注免疫 BAL B/ c鼠 ,5周和 10周后 ,分别取免疫鼠的肺、心脏、肝脏、脾、肌肉、肾脏及血液 ,抽提组织 DNA,并以此为模板进行 PCR扩增 ,扩增的产物经琼脂糖凝胶电泳分析。结果 免疫后 5周 ,p BK- P30免疫组的上述各器官组织 DNA中均扩增出 P30基因片段 ,其大小 10 2 7bp,p BK- p30 & p BK- P2 2免疫组分别扩增出 P30和 P2 2基因片段 ,P2 2基因片段为 5 93bp,均与预期结果相符合。NS对照组和 p BK- CMV空质粒对照组未扩增出片段。免疫后 10周 ,仅免疫鼠血液扩增出上述特异性基因片段。结论 弓形虫 DNA疫苗免疫 BAL B/ c鼠 5周后 ,多个不同器官组织均可检测出重组质粒 ,免疫后 10周血液检测出重组质粒。 Aim\ Directly immunize BALB/c mice via intramuscular injection with DNA vaccines pBK-P30、pBK-P22,to observe the distribution of the recombinant plasmids in different organs and tissues of mice.Methods\ Recombinant plasmids pBK-P30、pBK-P22 were injected into the BALB/c mice via muscles ,after 5 weeks immunization,taking lung,heart liver,spleen,muscle and blood,extracting tissue DNA as templates,amplifying P30 and P22 gene fragments by means of polymerase chain reaction (PCR)and identifying the amplification products by agarose gel electrophoresis.Results\ P30 gene fragments were amplified out from those tissues of the pBK-P30 vaccinated group mice,P30 and P22 gene framents were amplified out from those tissues of the pBK-P30&pBK-P22 vaccinated group mice respectively,the amplified fragments size of P30 and P22 were 1027bp and 593bp respectively,which were in accordance with the expected ones.NS and pBK-CMV control group mice could not be amplified any fragment.But those specific gene fragments were only amplified specially from blood after 10 weeks immunization.Conclusion\ After 10 weeks immunization,the combinant plasmids distribute different organs and tissues of the BALB/c mice immunized by DNA vaccines of Toxoplasma gondii,but only blood was Detected out the recombinant plasmids.
