摘要
目的 为从多方面证实赖型钩体重组基因疫苗的免疫活性。方法 将赖型钩体重组基因多肽疫苗 (分子量6 8k Da)多点皮下注射免疫豚鼠。(以质粒载体 p T7- 7为阴性对照 ,全钩死疫苗 WCV为阳性对照 )取其脾细胞 ,用 3H- Td R法和MTT比色法分别测定特异性淋巴细胞转化试验 (L TT)的相对转化指数 (RPI)及 IL - 2 ,IL - 6活性。结果 1)重组基因疫苗免疫组特异性 L TT的 RPI为 2 .19± 0 .18明显高于 p T7- 7阴性对照 1.42± 0 .2 7(P<0 .0 0 5 ) ,与阳性对照 WCV无统计学差异 (P>0 .0 5 )。2 )基因疫苗免疫组 IL - 2 ,IL - 6活性分别为 34.8± 3.11,94.6± 6 .0 2测定值明显高于 p T7- 7阴性对照 2 0 .4± 3.0 5 ,6 1±6 .2 8(P<0 .0 0 5 ) ,与 WCV阳性对照无统计学差异 (P>0 .0 5 )。结论 1)钩体基因重组疫苗能使免疫豚鼠体内 Th1 、Th2 活化增强 ,有胸腺依赖性抗原性质 (TDAg) ,可激起体内强有力 T- B细胞协同效应的特异性体液免疫反应 ,确有增强免疫活性作用 ,是良好的免疫原。2 )重组钩体基因疫苗与阳性对照 WCV各实验值无统计学差异 ,提示二者免疫活动可能相当 ,但重组基因疫苗有毒副作用小的优势 。
Aim To identification the immunization activation of recombinant Leptospira gene vaccine from many-side Methods The guinea pigs were immunized with recombinant Leptospira gene vaccine [plasmid vector pT7-7 was negtive control,inactivated whole cell vaccine(WCV) was positive control].Then spleen cells were taken out.Particularity lymphocyte transformation test(LTT),IL-2 and IL-6 activation of these spleen cells were determined by MTT and 3H-TdR respectively.Results 1)The Relative transformation index of gene vaccine group was significance higher than pT7-7 group (vaccin group:219±018,pT7-7 group 142±027(P<0005);2)the activation of IL-2 and IL-6 from recombinant gene vaccine group waw significance stronger than pT7-7 group (vaccine group IL-2:348±311,IL-6:946±603, pT7-7 group IL-2:204±305,IL-6:67±628),(P<0005).Conclusion 1)The activation of Th 1 and Th 2 lymphocyte cell were increased in the guinea pigs with gene vaccine immunized.It suggested the recombinant Leptospira gene vaccine could elicit an extremely strong immunization effect of T-cell cooperate with B-cell and the gene vaccine was equal the WCV(P>005)but the gene vaccine sideeffect was small and the applying prospect was good.
