鬼臼苦素对卵巢癌耐药SKOV-3/DDP细胞株裸鼠移植瘤的抑制作用 被引量:2

Inhibiting effect of picropodophyllin on SKOV-3/DDP cell strain transplantation tumor of ovarian cancer in drug-resistant nude mice

摘要 目的:观察鬼臼苦素对上皮性卵巢癌耐药系SKOV-3/DDP细胞株裸鼠移植瘤的作用并对其机制进行评价。方法:建立30只人卵巢癌裸鼠移植瘤模型,裸鼠随机分成两组,溶剂对照组(生理盐水)和PPP(40 mg/kg)组各15只,每隔3天腹腔给药1次,18天后处死,测量裸鼠实验前后体重及肝肾功能,绘制生长曲线,计算抑瘤率,免疫组化法测定肿瘤内磷酸化胰岛素样生长因子-1受体(pIGF-1R)、微血管密度(MVD)。结果:PPP组抑瘤率为(40.17±9.13)%。PPP组血清丙氨酸转移酶、尿素氮、肌酐值与对照组比较差异无统计学意义(P>0.05)。PPP组移植瘤内pIGF-1R﹑MVD较生理盐水对照组均明显降低,差异有统计学意义(P<0.05)。结论:鬼臼苦素对卵巢癌耐药SKOV-3/DDP细胞株裸鼠移植瘤具有抑制生长作用,且对裸鼠肝肾功能没有明显的不良反应。鬼臼苦素可能有抑制肿瘤新生血管生成的作用。 Objective: To observe the effect of picropodophyllin on SKOV - 3/DDP cell strain transplantation tumor of epithelial ovarian cancer in drug - resistant nude mice, and evaluate the mechanism. Methods : The models of transplantation tumor of human ovarian cancer were established in 30 nude mice, then the nude mice were divided into solvent control group (normal saline) and PPP group (40 mg/kg), 15 nude mice in each group; the nude mice were treated with tranabdominal injection every three days, then they were killed after 18 days, the body wight, liver function, and renal function of the nude mice before and after experiment were measured, then growth curve was drawn, the tumor inhibition rate was calculated, immunohistochemical method was used to detect phosphorylation insulin - like growth factor - 1 receptor ( pIGF - 1R) in tumor and microvessel density (MVD) . Results: The tumor inhibition rate in PPP group was ( 40. 17 ± 9. 13 )%. There was no statistically significant difference in serum alanine aminotransferase, urea nitrogen, and ereatinine levels between PPP group and control group (P 〉 0. 05 ) . pIGF - 1R and MVD in PPP group were statistically significantly lower than those in control group (P 〈0.05) . Conclusion: Picropodophyllin can inhibit the growth of SKOV -3/DDP cell strain transplantation tumor of ovarian cancer in drug - resistant nude mice, and it has no obvious adverse impact on liver function and renal function of nuder mice. Picropodophyl- lin may inhibit tumor angiogenesis.
