摘要
目的研究T一2毒素对低硒(Se)大鼠关节软骨抗氧化酶及基因表达的影响。方法新生断乳雄性健康sD大鼠,平均体重60-80g,随机分成两大组即常规组和低se组喂养30d,低Se动物模型建造成功后再分成常规组、低se组、常规+低T一2组、常规+高T.2组、低se+低T一2组、低se+高T一2组,再T~2毒索灌胃饲养30d后,观察低se、T一2毒素及二者共同作用对各组大鼠关节软骨丙二醛(MDA)含量、总抗氧化能力(T—AOC)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH—Px)活性及SOD、CAT、GSH—PxmRNA表达的影响。结果与常规组相比,各组大鼠关节软骨中MDA含量升高,T—AOC、SOD、CAT和GSH—Px活性下降,均有统计学意义(P〈0.05);低Se+低T一2组、低se+高T一2组与低se组相比,MDA含量升高,T—AoC、SOD、CAT和GSH—Px活性下降(P〈0.05);与常规组相比,各组大鼠关节软骨SOD、CAT、GSH—PxmRNA表达降低(P〈0.05)。结论低se或单纯T一2毒素均可减弱大鼠关节软骨抗氧化酶活性,低se与T.2毒素对机体抗氧化功能有协同作用。
Objective To study the effect of T-2 toxin on articular cartilage antioxidant enzymes and gene expression in selenium (Se) dificient rats. Method Weanling male SD rats (60-80) g were randomly divided into normal group and Se-deficient group and fed for 30 d. After the animal model of Se-deficiency was developed, the rats were randomly divided into 6 groups: normal group ( I ), Se-deficient group (II), normal+low T-2 group (III), normal+high T-2 group (IV), Se-deficient+low T-2 group (V), Se-deficient+high T-2 group (VI). The group I ,III, V were fed normal feed, and the other groups were fed low-Se feed. All groups were given T-2 toxin at 100 or 200 ng/(g bw'd) except the group I and II. At the 30th day of the experiment, the content of malondialdehyde (MDA), the total antioxidant capacity (T-AOC) and activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase(GSH-Px) and SOD, CAT, GSH-Px mRNA expression were observed. Results The MDA content in the other groups was significantly higher than group I (P〈0.05). Besides, the T-AOC and activities of SOD, CAT, GSH-Px in the other groups were significantly lower than group I (P〈0.05). Compared with group II, the content of MDA in group V and VI was increased and the T-AOC, activities of SOD, CAT, GSH-Px were decreased (P〈0.05) . Compared with group I, the mRNA expression of SOD, CAT, GSH-Px in the other groups were significantly lower (P〈0.05) . Conclusion Se-deficiency or T-2 toxin could weaken the activities of articular cartilage antioxidant enzymes. Se-deficiency and T-2 toxin had synergistic effect on the antioxidant function.
